The women's 4x100 metre freestyle relay of Swimming 2016 during the 2016 Summer Olympics was held on 6 August 2016 in the Rio de Janeiro Olympic Aquatics Stadium. With 16 teams competing, the event was won by the team from Australia, with United States taking the silver medal and Canada taking bronze.
This competition exists out of two rounds, each with eight teams. The eight teams with the fastest times in either semifinal advance to the final. The fastest team in the final is the winner of the event.

Sixteen teams qualified for the event, with Australia as the absolute favorite. As the defending Olympic champion, reigning world champion, and world record holders, the team had mostly dominated the event in the last few years. There were quite a few countries that would contend for the medals, including the United States, Canada, and the Netherlands. Italy and Sweden had very strong individual swimmers, such as Federica Pellegrini and Sarah Sjoestroem, and even Japan could surprise. Other outsiders included Denmark with Pernille Blume, Russia, and France.
